A pure noncompetitive inhibitor binds equally well to free enzyme and the enzyme-substrate complex at a site separate from the active site. The educational model uses effective Vmax = Vmax ÷ (1 + [I] ÷ Ki), while Km remains constant. The molecular scale and animation time are conceptual.

Applications and misconceptions

Noncompetitive and mixed inhibitors are important in drug discovery, toxicology, metabolic control, and enzyme assays. An allosteric inhibitor need not block substrate binding directly. Extra substrate cannot overcome pure noncompetitive inhibition, and real inhibitors often show mixed rather than perfectly pure behavior.
